Is it possible on an AcornCNC to output step and direction
commands using the screws terminals for axis 1 and 2 and
the DB25 terminals for axis 3 and 4?

Screw terminals are for servo drives, and the DB25 is for 5V stepper drives. If you have servos and steppers, you can use both.

I wired my machine to use axis 1 and 2 with Yaskawa Sigma drive. These are working nicely. I've wired a very small steppers drive to axis 3 using the 6 (step) and 7 (direction) pins of the DB25. Custom DB25 settings is one but I didn't make any changes to it.
As soon as I command axis 3 to move, the green enable LED turns to red.
What have I done wrong?

I unchecked the custom DB25 settings switch and relaunched. All of the axis turn the green enable LED to red.
I reselected Sigma Yaskawa settings in Axis Motor Drive Configuration. Relaunched. Now all axis are working. The green enable lights stay green.
Happy to have made progress. Curious to know what happened in the background that solved my problem.
